Toro Magazine has an interview with Canadian actress Cindy Busby, who apparently is in The Vampire Diaries pilot. Could this be the unidentified actress playing Brooke? Sure looks like her – and she confirms in the blog on her official website:

Last March I shot the pilot and first episode to the CW’s new show “The Vampire Diaries”… I’m so excited because I get to open up the series (meaning I’m the first actor you ever see in the show)… AWESOME!!!

Busby plays Ashley Stanton, a regular, on the CBC TV series Heartland. She’s also “a serious vampire fan.”

“I personally have always been a huge vampire fan. When I was a kid, it was all that I would ever have nightmares about, which is so weird. But at the same time, I enjoyed it. It was kind of like an enjoyable torture.”

Back in April, she wrote a brief post about her experience filming The Vampire Diaries pilot:

I got to realize a life long dream of being in a Vampire project. This week I shot some scenes in the new CW pilot called “Vampire Diaries” produced by the legendary Kevin Williamson! I was SOOO excited to meet and work with him. Mr. Williamson also gave me the best compliment… after hearing my famous scream, he said that I reminded him of a young Jamie Lee Curtis…. holy smokes, my jaw dropped! Everyone on the set was amazing and friendly and extremely professional, which is always such an enjoyable experience! I have a wonderful feeling that this show will be a hit!
